本発明において、生理活性物質として細胞傷害性物質、低分子抗ウイルス剤等の非極性物質或いは極性基を多く有しない物質を使用する場合、(1)小胞体内部に内包する或いは(2)膜構成成分と疎水結合させて膜中に入れるか、のいずれかの方法が用いられる。これらの場合は静電反発を利用して小胞体同士の凝集を防止する目的で、アニオン性を有する物質を使用することができる。アニオン性を有する物質としては、好ましくは酸性リン脂質を例示することができる。
酸性リン脂質としては、ホスファチジルグリセロール、ホスファチジルセリン、ホスファチジルイノシトール、メトキシポリエチレングリコール付加ホスファチジルエタノールアミン、及びホスファチジン酸を例示することができる。
なお本明細書において引用された全ての先行技術文献は、参照として本明細書に組み入れられる。
〔実施例1〕
N-(α-トリメチルアンモニオアセチル)-ジオレイル-D-グルタメート クロリド (相互薬工株式会社、日本;製品名:DC-3-18:1)、ジオレオイルホスファチジルエタノールアミン(日油株式会社、日本)及びコレステロール(和光純薬株式会社、日本)を、配合比40:30:30(モル比)にて適量のクロロホルムに溶解させ、これを減圧下に溶媒を留去させながら乾固させて、脂質混合物とした。
50 mLバイアル中に、リポソーム分散液10 mLを入れ更にトコフェロール液400 μLを入れて(全脂質モル数に対して20 %)、ボルテックスにて20 秒間混合攪拌した。更に1時間室温にて1時間インキュベーションしてビタミンE吸着リポソーム分散液を得た。平均粒径は124.2 nmであった。
DL-α-トコフェロール(東京化成株式会社、日本)2.5 μmolに対して400 μLのエチルアルコールの割合で溶解させて、0.2 μmのフイルターでろ過した液(トコフェロール液)を調整した。
50 mLバイアル中に、〔実施例1〕で作製したリポソーム分散液10 mLを入れ更にトコフェロール液400 μLを入れて(全脂質モル数に対して10.0 %)、ボルテックスにて20秒間混合攪拌した。更に室温にて1時間インキュベーションしてビタミンE吸着リポソーム分散液を得た。平均粒径は136.7 nmであった。
DL-α-トコフェロール(東京化成株式会社、日本)3.75 μmolに対して400 μLのエチルアルコールの割合で溶解させて、0.2 μmのフイルターでろ過した液(トコフェロール液)を調整した。
50 mLバイアル中に、〔実施例1〕で作製したリポソーム分散液10 mLを入れ更にトコフェロール液400 μLを入れて(全脂質モル数に対して15.0 %)、ボルテックスにて20 秒間混合攪拌した。更に1時間室温にて1時間インキュベーションしてビタミンE吸着リポソーム分散液を得た。平均粒径は133.5 nmであった。
N-(α-トリメチルアンモニオアセチル)-ジオレイル-D-グルタメート クロリド (相互薬工株式会社、日本;製品名:DC-3-18:1D)、ジオレオイルホスファチジルエタノールアミン(日油株式会社、日本)及びコレステロール(和光純薬株式会社、日本)を配合比40:30:30(モル比)にて適量のクロロホルムに溶解させ、これを減圧下に溶媒を留去させながら乾固させて、脂質混合物とした。溶媒が確実に除去できる間でデシケーターにて乾燥させた後、この脂質混合物に9%スクロース溶液を加え、65℃加温下、ソニケーターで間接的に超音波照射することで全体の脂質濃度2.5mMのリポソーム粗分散液を得た。次に、リポソームの粒子径を揃えるため、孔径0.2μmのフイルターを2枚重ねてエクストル―ダーにセットし、約65℃加温・加圧下にエクトルージョン(押し出しろ過)した。更に孔径0.1μmのフイルターを2枚用いて同様にエクトルージョンし、これを空のリポソーム分散液とした。平均粒径は138.7nmであった。
次にNano-Mizer mark II NM2-L200(吉田機械興業株式会社、日本)にて、100MPaの加圧下に3回通過させて更なる微粒子化処理を行った結果、平均粒径は55.7nmであった。
50mLバイアル中に、リポソーム分散液10mLを入れ更にトコフェロール液320μLを入れて(全脂質モル数に対して16%)、ボルテックスにて20秒間混合攪拌した。更に1時間室温にて1時間インキュベーションしてビタミンE吸着リポソーム分散液を得た。平均粒径は58.0nmであった。
〔実施例1〕で作製した平均粒径136.1 nmのリポソーム分散液をそのまま用いた(トコフェロール無添加)。
〔比較例2〕
DL-α-トコフェロール(東京化成株式会社、日本)20 μmolを400 μLのエチルアルコールに溶解させて、0.2 μmのフイルターでろ過した液(トコフェロール液)を調整した。
なお、実施例はこれだけに留まるわけではなく、一例に過ぎないことを明記しておく。
C型肝炎は、日本国内で200万人、世界的には2億人以上にものぼる感染者が存在する、重大な感染症の一つである。その大きな特徴は、C型肝炎ウイルス(HCV)がヒトに感染すると、80~90%の高率で持続感染化し、慢性肝炎、さらには肝細胞がんを引き起こすことにある。現行のペグインターフェロン・リバビリン併用療法により治療成績の向上をみたものの、未だに治療効果は十分とは言えず、より有効で副作用の少ない治療の開発が急務となっている。そこで、短い二本鎖RNA (siRNA)によるHCVの制御を試みた。
本実施例では、ルシフェラーゼ遺伝子を含むHCVレプリコン細胞としてR6 FLR41-14細胞(Watanabe, T. et al., Gene Theraphy 13: 883-892 (2006))を用いて、siRNAアッセイを行った。各々の細胞の細胞数を数え、10% 非働化済FCSを含むDMEM+GlutaMax-Iにけん濁し、R6 FLR41-14細胞は4500cells/100 μL/wellで96wellにまき、翌日にsiRNAのトランスフェクションを行った。siRNAはsi197-1(センス鎖5'- AUCAUGAGCACAAAUCCUAAA -3'、配列番号:1、アンチセンス鎖(3'- CGUAGUACUCGUGUUUAGGAU -5'、配列番号:2)を使用した。siRNAの終濃度は、0.01、0.04、0.11、0.33、1、1.1、3、3.3、10、30、90 nMとした。
それぞれの、コンジュゲート前後の粒径とゼータ電位を表1に示した。
ルシフェラーゼ活性測定においては、Bright-Glo Luciferase Assay System (Promega, cat.#E2620)を使用した。方法は論文(Watanabe, T. et al., Gene Therapy 13: 883-892 (2006))にしたがった。
ルシフェラーゼアッセイ用96ウェルプレートの全ウェルの培地を廃棄し、5% 非働化済FCSを含むDMEM+GlutaMax-Iを75 μL/wellで加え、培地交換した。Bright-Glo Luciferase Assay Systemを75μL/wellで加えて、Mithras LB 940(Berthold)で1分間振とうした後、ルシフェラーゼ活性を測定した。
細胞毒性測定においては、Cell Counting Kit-8(Dojindo, cat.#343-07623)を使用した。
Cell Counting Kit-8溶液を、5%非働化済FCSを含むDMEM+GlutaMax-Iで7%に希釈し、アッセイ用の溶液を作製した。細胞毒性試験用の96ウェルプレートの全ウェルの培地を廃棄し、上述のアッセイ用の溶液を100 μL/wellで加え、37℃、5% CO2にて1時間インキュベートした。マイクロプレートリーダー(Bio-Rad model 550)を用いて、波長450 nm(参照波長655 nm)を測定した。
その結果トコフェロールを付加した〔実施例1〕及び〔実施例2〕で製造のリポソーム分散液は強力なHCVレプリコン複製阻害活性を示し、細胞毒性については軽微であることが明らかになった。〔比較例1〕のリポソーム分散液に比較して〔実施例2〕で製造のリポソームは約100倍、〔実施例1〕で製造のリポソーム分散液は約1000倍のHCVレプリコン複製阻害活性を示した(図1、図2)。
〔動物試験実施例1〕デザインしたsiRNAのHCV遺伝子導入マウスでの阻害活性の検討
<HCV遺伝子導入マウス>
HCVの持続感染成立機序、慢性肝炎・肝硬変・肝癌への推移機構解明の為に、病態モデルのトランスジェニック( Tg )マウス作製が行われてきた。HCV感染により近いモデルとして、任意の時期にHCV遺伝子をスイッチング発現することができるCre/loxP recombinant systemを用いたHCV構造蛋白質領域発現Tgマウス ( CN2 マウス )が樹立された。これまではスイッチングの方法として、Cre DNA recombinaseを発現する組換えアデノウイルス(AxCANCre)の静脈内投与が行われてきた。しかしAxCANCre法では、マウスのアデノウイルスに対する免疫応答も惹起されてしまう。そこで本発明者らはCre/loxPシステムでHCV遺伝子を導入したトランスジェニックマウスとCreを誘導するMx-Creトランスジェニックマウスを交配することにより、任意の時期にHCV遺伝子をスイッチング発現するトランスジェニック(Cre/loxP/HCV-MxCre Tg)マウスを作製した。MxCre Tg マウスでは、Cre DNA recombinaseがインターフェロン(IFN)反応性 Mx1プロモーターによって発現される為、poly-IpC(以下pIpCと記載する、二重鎖 RNA)の投与によるIFNの誘導によってスイッチングが起こる。このマウスにpIpCを1日置きに3回腹腔内投与した。初回投与後0.5日目からHCV コア蛋白質の発現が認められ、7日目で915 pg/mg(肝総蛋白質量)とピークに達した。その後、減少傾向にあるものの、21日目でも435 pg/mgと発現が持続されている。さらに、この発現は6ヶ月を過ぎても同レベルで維持されていた(図3、図4)。
遺伝子発現43日後のCre/loxP/HCV-MxCre Tgマウスに〔実施例1〕で製造したリポソーム分散液とsi197-1複合体(0.3mg siRNA/kg体重)を投与して、そのRNAi活性を評価した。投与後2日目に剖検し、肝臓組織のサンプリングを行った。この肝臓中のHCVコア蛋白質量をELISA法(オーソ HCV コア蛋白 ELISA キット)にて定量し、RNAi活性を評価した。絶食、非絶食群ともにsiCTRL (ダーマコン社; siCTRL3)投与群に比較して顕著なHCVコア蛋白質量の低下が認められた(図5)。これは、これまでに報告された物(Watanabe, T. et al., J. Hepatology 47: 744-750 (2007))に比較して50倍以上強い阻害活性を示すことが明らかとなった。

- 前記カチオン性ポリマーが、ポリエチレンイミン又はポリリジンであることを特徴とする請求項7に記載の小胞体。
- 前記アニオン性を有する物質が、酸性リン脂質であることを特徴とする請求項1に記載の小胞体。
- 前記酸性リン脂質が、ホスファチジルグリセロール、ホスファチジルセリン、ホスファチジルイノシトール、メトキシポリエチレングリコール付加ホスファチジルエタノールアミン、及びホスファチジン酸からなる群から選択される少なくとも1種類以上を含むことを特徴とする、請求項9に記載の小胞体。

- 前記ビタミンEが、dl-α-トコフェロールであることを特徴とする請求項1に記載の小胞体。
- 前記カチオン性を有する物質が、小胞体を構成する物質全体のモル数の10~50%の範囲で含まれていることを特徴とする請求項1~13のいずれかに記載の小胞体。
- 前記ビタミンEが、小胞体を構成する物質全体のモル数の、3~30%の範囲で吸着又は化学結合されていることを特徴とする請求項1~14のいずれかに記載の小胞体。
- 前記ビタミンEが、小胞体を構成する物質全体のモル数の、5~20%の範囲で吸着又は化学結合されていることを特徴とする請求項1~14のいずれかに記載の小胞体。
- 前記小胞体の粒径が50~150nmの範囲であることを特徴とする、請求項1~16のいずれかに記載の小胞体。
- プラスミドDNA、siRNA、miRNA、アンチセンスオリゴヌクレオチド、抗ウイルス剤、及び細胞傷害性物質からなる群から選択される少なくとも1種類以上の生理活性物質を小胞体が内封する、及び/又は該生理活性物質が小胞体の成分に疎水的又は静電的に結合することを特徴とする、請求項1~17のいずれかに記載の小胞体。
- 小胞体表面のゼータ電位がアニオニックであることを特徴とする請求項18に記載の小胞体。
- 前記生理活性物質が、抗ウイルス剤であることを特徴とする請求項18に記載の小胞体。
- 前記細胞傷害性物質が、代謝拮抗剤、プラチナ系剤、アルキル化剤、植物アルカロイド、分子標的薬、生物学的応答調節剤、及び抗がん性抗生物質からなる群から選択される少なくとも1種類以上を含むことを特徴とする、請求項18に記載の小胞体。
- 前記代謝拮抗剤が、エノシタビン、カペシタビン、カルモフール、クラドリビン、ゲムシタビン、シタラビン、シタラビンオクホスファート、テガフール、テガフール・ウラシル、テガフール・ギメラシル・オテラシルカリウム、ドキシフルリジン、ネララビン、ヒドロキシカルバミド、フルオロウラシル、フルダラビン、ペメトレキセド、ペントスタチン、メルカプトプリン、及びメトトレキサートからなる群から選択され、
前記プラチナ製剤が、オキサリプラチン、カルボプラチン、シスプラチン、及びネダプラチンからなる群から選択され、
前記アルキル化剤が、イホスファミド、シクロホスファミド、ダカルバジン、テモゾロミド、ニムスチン、ブスルファン、メルファラン、及びラニムスチンからなる群から選択され、
前記植物アルカロイドが、イリノテカン、エトポシド、ソブゾキサン、ドセタキセル、ノギテカン、パクリタキセル、ビノレルビン、ビンクリスチン、ビンデシン、ビンブラスチンから選ばれるものであり、分子標的薬がイマチニブ、エルロチニブ、スニチニブ、セツキシマブ、ソラフェニブ、パニツムマブ、ベバシズマブ、ボルテゾミブからなる群から選択され、
前記生物学的応答調節剤が、インターフェロン-α、インターフェロン-β、インターフェロン-γ、インターロイキンからなる群から選択され、
前記抗がん性抗生物質が、アクラルビシン、エピルビシン、ダウノルビシン、ドキソルビシン、ピラルビシン、ブレオマイシン、マイトマイシンC、及びミトキサントロンからなる群から選択されることを特徴とする、請求項21に記載の小胞体。 - 請求項18~22のいずれかに記載の小胞体、及び薬学的に許容される担体を含む、生理活性物質を血清存在下で肝実質細胞に送達するために用いられるキット。
- 請求項18~22のいずれかに記載の小胞体を、対象に投与する工程を含む、対象において生理活性物質を肝実質細胞に送達する方法。
