Description
本発明は鼻腔内投与により嗅上皮から嗅球を経て中枢神経系組織を標識するための経鼻中枢神経系組織標識用組成物、経鼻中枢神経系組織標識用組成物を用いた標識方法、及び経鼻中枢神経系組織標識用組成物を用いたスクリーニング方法に関する。
中枢神経系組織標識用組成物は、生体の中枢神経系組織に移行性を持ち、中枢神経系組織内の特定の部位に偏在する性質を有する化合物である。外部から適当な計測手段を用いて、中枢神経系組織標識用化合物の偏在状態を可視化することにより、生体中枢神経系組織の活動を細胞・分子レベルで観察することや、中枢神経系組織疾患を診断することが可能となる。
フッ素18標識フルオロデオキシグルコース(FDG)は、従来、中枢神経系組織標識用に用いられている化合物の一つであり、中枢神経系組織内の脳腫瘍への偏在状態を陽電子放射断層撮影法(PET)により可視化することにより、脳腫瘍の診断に役立つ情報を取得することができる。
特許文献1に開示されている化合物は、アミロイドβ蛋白質に親和性を有する近赤外蛍光色素であり、近赤外蛍光計測手段を用いてアミロイドβ蛋白質の沈着した中枢神経系組織部位を可視化することにより、アルツハイマー病の診断に役立つ情報を取得することができる。
ところで、特許文献2には、ゼブラフィッシュを用いた中枢神経系に対する化合物スクリーニングの方法が開示されている。同文献によれば幼若なゼブラフィッシュは染料化合物の脳への移行が確認されるが、受精後10日目には、おそらく血液脳関門が形成されるために、中枢神経系への化合物の移行が見られなくなるとの記載がある。
脳への化合物の移行性は血液脳関門(BBB)や、血液脳脊髄液関門(BCSFB)によって制限されており、通常組織であれば組織内に移行する化合物であっても脳内へ移行できないものが多い。そのため、BBBやBCSFBの影響を受けずに中枢神経系組織を明瞭に標識可能な中枢神経系組織の標識用化合物が求められていた。
また、従来知られた中枢神経系組織標識用組成物は通常、皮内、腹腔内、静脈、動脈、または脊髄液への注射により投与される。中枢神経系組織以外の全身の組織に対しても循環系により分配される化学物質は、アナフィラキシーショック、心不全、肝機能障害、腎機能障害、皮膚障害、呼吸器障害等の安全面を考慮して開発をする必要があった。
本発明が解決しようとする課題は、全身への移行性が少なく、より生体への影響の少ない投与経路での使用が可能な中枢神経系組織標識用組成物を提供することである。
前記課題を克服するため、本発明者らは鋭意検討の結果、循環系を経て全身に移行する割合が比較的に少ない、鼻腔内投与により嗅上皮から嗅球を経て中枢神経系組織を標識することができる経鼻中枢神経系組織標識用組成物を見出して本発明に至った。
すなわち本発明は、鼻腔内投与により生体の嗅上皮から嗅球を経て中枢神経系組織を標識するための経鼻中枢神経系組織標識用組成物であって、下記一般式(1)、または(2)のいずれかの化学式で表わされる化合物を少なくとも1種を有効成分として含むことを特徴とする経鼻中枢神経系組織標識用組成物。
(一般式(1)中、R1は、アルキル基を表し、R3~R6は、各々独立して水素原子、アルキル基、アリール基、アルコキシ基、カルボン酸基、スルホン酸基、ヘテロ環基、アミノ基、またはハロゲン原子を表し、R5とR6は互いに結合して環を形成しても良い。X1 -は陰イオン性基を表す。Aは下記一般式(3)または(4)を表す。)
本発明の経鼻中枢神経系組織標識用組成物は、鼻腔内投与により嗅上皮から嗅球を経て中枢神経系組織を標識する。そのため、全身の組織に対して循環系により分配されることに起因する生体への影響を低減しながら、簡便且つ高精細に中枢神経系組織の形態や細胞状態などを評価・解析することが可能となる。
また、本発明の経鼻中枢神経系組織標識用組成物は、鼻腔内投与により経時的に嗅上皮から嗅球を経て中枢神経系組織へ移行していくため、その過程での標識用組成物の偏在状態をモニターすることにより、嗅覚神経細胞の繋がりを可視化することが可能となる。
更に、ある化合物の中枢神経系への移行メカニズムが本発明の経鼻中枢神経系組織標識用組成物の中枢神経系組織への移行メカニズムと拮抗するような場合、本発明の経鼻中枢神経系組織標識過程は、その化合物との共存により、影響を受けることになる。この影響を検出することにより、その化合物が経鼻投与により中枢神経系組織へ移行する可能性を見出すことが出来る。すなわち本発明の経鼻中枢神経系組織標識用組成物を用いれば、経鼻投与により中枢神経系組織へ移行する薬剤をスクリーニングすることが可能となる。
以下、本発明を具体的な実施形態により説明するが、ここで挙げる実施形態は、経鼻中枢神経系組織標識用組成物の一例に過ぎず、本発明はそれらの実施形態によって限定されない。
本発明の第一実施形態である経鼻中枢神経系組織標識用組成物は、鼻腔内投与により生体の嗅上皮から嗅球を経て中枢神経系組織を標識するための経鼻中枢神経系組織標識用組成物であって、下記一般式(1)または(2)のいずれかの化学式で表わされる化合物を少なくとも1種を有効成分として含むことを特徴とする。
一般式(1)中、R1は、アルキル基を表し、R3~R6は、各々独立して水素原子、アルキル基、アリール基、アルコキシ基、カルボン酸基、スルホン酸基、ヘテロ環基、アミノ基、またはハロゲン原子を表し、R5とR6は互いに結合して環を形成しても良い。X1 -は陰イオン性基を表す。Aは下記一般式(3)または(4)を表す。
一般式(3)中、R2は、アルキル基を表し、R7~R10は、各々独立して水素原子、アルキル基、アリール基、アルコキシ基、カルボン酸基、スルホン酸基、ヘテロ環基、アミノ基、またはハロゲン原子を表し、R7とR8は互いに結合して環を形成しても良い。
一般式(4)中、Yはアルキル基を表す。*は結合部位を示す。
前記一般式(1)及び(3)中のR1~R2におけるアルキル基としては、特に限定されるものではないが、例えば、メチル基、エチル基、プロピル基、ブチル基等が挙げられる。さらに、該基には、本発明の色素化合物の保存安定性を著しく阻害するものでなければさらに置換基を有していてもよい。
R3~R10におけるアルキル基としては、特に限定されるものではないが、例えば、メチル基、エチル基、プロピル基、ブチル基等が挙げられる。
R3~R10におけるアリール基としては、特に限定されるものではないが、例えば、フェニル基、ナフチル基などが挙げられる。
R3~R10におけるアルコキシ基としては、特に限定されるものではないが、例えば、メトキシ基、エトキシ基、プロポキシ基、ブトキシ基等が挙げられる。
R3~R10におけるヘテロ環基としては、特に限定されるものではないが、ピリジル基、ピラジル基、モルホリニル基等が挙げられる。
R3~R10におけるアミノ基としては、特に限定されるものではないが、例えば、無置換アミノ基;N-メチルアミノ基、N-エチルアミノ基等のモノ置換アミノ基;N,N-ジメチルアミノ基、N,N-ジエチルアミノ基、N,N-メチルプロピルアミノ基等のジ置換アミノ基が挙げられる。
R3~R10におけるハロゲン原子としては、例えば、フッ素原子、塩素原子、臭素原子、または、ヨウ素原子等が挙げられる。
R5とR6、R7とR8は互いに結合して形成される環としてはフェニル基等が挙げられる。
X1 -における陰イオン性基としては、特に限定はされないが、例えば、塩化物イオン、臭化物イオン、ヨウ化物イオン等のハロゲンイオン、硫酸イオン、リン酸イオン等の無機酸イオン、酢酸イオン等の有機酸イオン等が挙げられる。

本発明にかかる一般式(1)、または(2)で表される色素化合物は、市販されており入手可能であるが、公知の方法によって合成することも可能である。
一般式(1)、または(2)で表わされる色素化合物の好ましい具体例は次に示す式(5)~(7)に示すが、これらに限定されるわけではない。
本発明の経鼻中枢神経系組織標識用組成物に含まれる化合物の濃度は中枢神経系組織を検出することが出来れば特に限定されるものではないが、標的とする部位や使用される化合物によって適宜調節される。通常は0.001ng/mL以上、100μg/mL以下の濃度で用いられ、より好ましくは0.001ng/mL以上、10μg/mL以下、より好ましくは0.001ng/mL以上、5μg/mL以下の濃度で用いられる。
本発明の経鼻中枢神経系組織標識用組成物は、前記一般式(1)、または(2)で表される化合物の少なくとも1種を適当な溶媒に溶解させて用いる。生体に影響がなければ、特に限定されるものではないが、例えば、生体との親和性が高い水性の液体が好ましい。具体的には、水;生理食塩水;リン酸緩衝液(PBS)、Tris等の緩衝液;メタノール、エタノール、イソプロパノール、ブタノール、エチレングリコール、グリセリン等のアルコール系溶媒;N,N-ジメチルスルホキシド(以下、「DMSO」と略する)、N,N-ジメチルホルムアミド(以下、「DMF」と略する)等の有機溶媒;D-MEM(Dulbecco’s Modified Eagle Medium)、HBSS(Hanks' Balanced Salt Solutions、ハンクス平衡塩)等の細胞培養用培地、または乳酸リンゲル液等の輸液が挙げられる。特に水が50%以上含まれていることが好ましい。また、これらの溶媒を2種以上混合して用いることも出来る。
本発明の経鼻中枢神経系組織標識用組成物の作製方法は、特に限定されるものではないが、例えば、上記のような溶媒に溶解させた化合物の濃厚溶液を希釈して作成しても良い。化合物の水溶性が低い場合には、適当な溶媒に溶解させてから精製水に希釈させて用いることが出来る。特に好ましくは、メタノール、エタノール、DMSO等である。
本発明の経鼻中枢神経系組織標識用組成物には、生体に適した塩濃度やpH等を制御することが必要であれば添加剤を1種類またはそれ以上組み合わせて添加することが出来る。
本発明の経鼻中枢神経系組織標識用組成物には、生体に適した塩濃度やpH等を制御することが必要であれば添加剤を1種類またはそれ以上組み合わせて添加することが出来る。
本発明に用いられる添加剤としては、経鼻中枢神経系組織標識用組成物に影響がなければ特に限定されるものではないが、例えば、保湿剤、表面張力調製剤、増粘剤、塩化ナトリウムのような塩類、各種pH調製剤、pH緩衝剤、防腐剤、抗菌剤、甘味剤、または香料等が挙げられる。
pH調製剤としては、pHを5~9に調製することが好ましく、特に限定されるものではないが、例えば、塩酸、酢酸、リン酸、クエン酸、リンゴ酸、水酸化ナトリウム、または炭酸水素ナトリウム等が挙げられる。
(標識対象)
本発明の経鼻中枢神経系組織標識用組成物を用いて嗅上皮から嗅球を経て中枢神経系組織を標識することが可能な生物種としては、特に限定されるものではないが、例えば、脊椎動物としては、トラフグ、クサフグ、ミドリフグ、メダカ、ゼブラフィッシュ等の硬骨魚類、アフリカツメガエル等の両生類、ニワトリ、ウズラ等の鳥類、ラット、マウス、ハムスター等の小動物、ヤギ、ブタ、イヌ、ネコ、ウシ、ウマ等の大動物、サル、チンパンジー、ヒト等の霊長類が挙げられる。特に、これらの生物個体の中枢神経系組織を生きたままの状態で染色することが出来る。また、生物種としては、ヒトを除外してもよい。

本発明の経鼻中枢神経系組織標識用組成物を用いて標識できる中枢神経系組織は、例えば、大脳、中脳、小脳、間脳、延髄、脊髄、視索、上丘(視蓋)、脳下垂体、視蓋脊髄(延髄)路、網様体などから構成される中枢神経系組織、これら組織の疾患状態組織、または疾患による新生組織及び癌組織等が挙げられる。又、生物種類、発生段階、または発生異常、疾病等によって前記と異なる中枢神経系組織が存在する場合は、それらの組織も含むことが出来る。
前記中枢神経系組織に含まれる細胞としては特に限定されるものではないが、神経細胞、オリゴデンドロサイト、シュワン細胞、プルキンエ細胞、アマクリン細胞、網膜神経節細胞、錐体細胞、星状細胞、顆粒細胞、グリア細胞、またはこれらの腫瘍細胞、これらの未分化状態の細胞(幹細胞)等が挙げられる。
本発明の第二の実施形態である投与経路は、生体の鼻腔内に上記の経鼻中枢神経系組織標識用組成物を投与することを特徴とする。本発明の経鼻中枢神経系組織標識用組成物は、生体の鼻腔内に投与することにより、経時的に嗅上皮から嗅球を経て中枢神経系組織を標識することができる。すなわち、投与直後には嗅覚受容細胞が標識され、次に嗅球が標識され、更には大脳(終脳)が標識される。鼻腔内投与後の経過時間により、標識部位を識別して細胞形態を染色することが出来る。
生体の鼻腔内への投与方法としては、特に限定されるものではないが、例えば、鼻腔内への噴霧または塗布等の手段による鼻粘膜への接触による方法等が挙げられる。
動物への投与の場合には、その投与形態、投与量は対象となる動物の体重や状態によって適宜選択する。

本発明の第三の実施形態の標識の可視化による情報の取得方法は、経鼻中枢神経系組織標識用組成物を用いて生体の嗅上皮から嗅球を経て中枢神経系組織を染色する過程を含むことを特徴とする。つまり、本発明の経鼻中枢神経系組織標識用組成物を鼻腔内に投与した後、一定時間後に、励起波長の光を観察部位に照射し、発生するより長波長の蛍光を観測し画像を形成することを特徴とする。
また、本発明の中枢神経系組織標識用組成物は、嗅神経をはじめとする脳神経も標識できる。本発明の好ましい標識の具体的方法には、染色や放射性核種で標識されたプローブ等を用いる方法がある。脳神経を染色できることで、中枢神経系組織とつながる末梢神経系組織の分布や配向などをイメージングすることができるため、好ましい。
本発明において、中枢神経系組織の細胞形態を染色するとは、中枢神経系組織に存在する細胞が少なくとも1種類以上染色され、1種類毎に細胞形態をたとえば蛍光等によってはっきりと判別することが可能な状態になることである。
本発明の経鼻中枢神経系組織標識用組成物は、蛍光性を有する蛍光性化合物であることが好ましい。前記式(5)~(7)もこの範疇に含まれる。本発明の経鼻中枢神経系組織標識用組成物を用いた嗅覚神経回路または中枢神経系組織の標識方法は、本発明の経鼻中枢神経系組織標識用組成物を鼻腔内に投与した後、一定時間後に、励起波長の光を観察部位に照射し、発生する励起波長より長波長の蛍光を観測し画像を形成することにより、標識部位の細胞形態情報を取得することができる。
本発明で用いられる観察方法は、中枢神経系組織に影響を与えなければ特に限定されるものではないが、生物試料の状態及び変化を画像として捉える方法である。例えば、中枢神経系組織に可視光、近赤外光、または赤外光を照射してカメラやCCD等で観察する可視光観察、近赤外光観察、赤外光観察、レーザー顕微鏡観察、または蛍光内視鏡等のように生物試料に対して励起光光源から励起光を照射して、発光している生物試料の蛍光を観察する蛍光観察、蛍光顕微鏡観察、蛍光内視鏡観察、共焦点蛍光顕微鏡観察、多光子励起蛍光顕微鏡観察、狭帯域光観察、または光干渉断層画像観察(OCT)、更に、軟エックス線顕微鏡による観察等が挙げられる。
本発明で用いられる励起するための波長は、特に限定されるものではないが、使用する前記一般式(1)、または(2)で表される色素化合物によって異なり、本発明の前記一般式(1)、または(2)で表される色素化合物が効率よく蛍光を発すれば特に限定されるものではない。通常、200~1010nm、好ましくは400~900nm、より好ましくは、480~800nmである。近赤外領域の光を用いる場合は、通常600~1000nmで、好ましくは、生体透過性に優れている680~900nmの波長が用いられる。
本発明で用いられる蛍光励起光源としては特に限定されるものではないが、各種レーザー光源を用いることが出来る。例えば、色素レーザー、半導体レーザー、イオンレーザー、ファイバーレーザー、ハロゲンランプ、キセノンランプ、またはタングステンランプ等が挙げられる。又、各種光学フィルターを用いて、好ましい励起波長を得たり、蛍光のみを検出したりすることが出来る。
このように生物個体に励起光を照射することにより、中枢神経系組織の内部において発光させた状態で中枢神経系組織を撮像すれば発光部位を容易に検出することが出来る。また、可視光を照射して得られた明視野画像と励起光を照射して得られた蛍光画像を画像処理手段で組み合わせることで、より詳細に中枢神経系組織を観察することも出来る。また、共焦点顕微鏡を用いれば、光学的な切片画像を取得することができるため、好ましい。さらに、多光子励起蛍光顕微鏡は、高い深部到達性と空間解像力を持つため、組織内部の観察に好ましく用いられる。
本発明の経鼻中枢神経系組織標識用組成物は放射性核種で標識されたプローブとして用いることも可能である。標識に用いる放射性核種の種類は、特に制限されるものではないが、使用の態様によって適宜選択することが出来る。具体的に、PETによる測定の場合は、例えば、11C、14C、13N、15O、18F、19F、62Cu、68Ga、または78Br等の陽電子放出核種を用いることが出来る。好ましくは、11C、13N、15O、または18F等であり、特に好ましくは、11C、または18F等である。また、SPECTによる測定の場合は、例えば、99mTc、111In、67Ga、201Tl、123I、または133Xe等のγ線放射核種を用いることが出来る。好ましくは、99mTc、または123I等である。更に、ヒト以外の動物を測定する場合には、例えば、125Iのようなより半減期の長い放射線核種を用いることが出来る。GREIによる測定の場合は、例えば、131I、85Sr、65Zn等を用いることが出来る。
放射性核種で標識した中枢神経系組織標識用組成物は、例えば、オートラジオグラフィー、陽電子(ポジトロン)放出核種を用いる放射断層撮影法(PET)、様々なガンマ線放出核種を用いる単一光子放射断層撮影法(SPECT)等によって画像化を行うことが出来る。また、フッ素原子核に由来するMR信号や13Cを利用した核磁気共鳴法(MRI)で検出してもよい。更に、次世代分子イメージング装置として複数分子同時イメージングが可能なコンプトンカメラ(GREI)等によって画像化することも可能である。また、例えば、液体シンチレーションカウンター、X線フィルム、イメージングプレート等を用いて中枢神経系組織用プローブの定量を行うことも可能である。
また、14C等の放射性同位元素で標識した経鼻中枢神経系組織用標識用組成物は、加速器質量分析法(AMS)等によって、血液中(または、尿中もしくは糞中)の濃度を測定して、標識化した物質の未変化体や代謝物の薬物動態学的情報(薬物血中濃度-時間曲線下面積(AUC)、血中濃度半減期(T1/2)、最高血中濃度(Cmax)、最高血中濃度到達時間(Tmax)、分布容積、初回通過効果、生物学的利用率、糞尿中排泄率等)を得ることが可能である。
放射線核種は一般式(1)、または(2)で表わされる化合物に含まれる形でも、結合する形でもよい。本発明の放射性核種を有するもしくは結合された化合物の具体例として、前記式(5)~(7)で表わされた化合物に、放射性核種が含まれた形の化合物、もしくは放射性核種が結合した形の化合物を例示することが出来る。
放射線核種の標識の方法は、特に限定されるものではなく、一般的に用いられている方法で良い。また、一般式(1)、または(2)で表わされる化合物を構成する元素の少なくとも一部を放射性核種で置換する形でも、結合する形でも良い。
一般式(1)、または(2)で表わされる化合物を放射性核種で標識した場合、1mMあたり、1~100μCi程度の放射性を有することが好ましい。
この時、用いる経鼻中枢神経系組織標識用組成物の投与量は、影響がなければ特に制限はされず、化合物の種類及び標識に用いた放射性核種の種類により適宜選択される。
本発明の第四の実施形態である情報の利用方法として、脳腫瘍/脳梗塞診断/脳内視鏡(ファイバースコープ)を用いた診断/治療/手術へ経鼻中枢神経系組織標識用組成物を利用することができる。
本発明の経鼻中枢神経系組織標識用組成物は、例えば、脳外科手術中に疾患の細胞組織や腫瘍と思われる被験物質の部位を特異・選択的に染色し、正常な細胞との違いを見極める手段や、疾患による組織の変化を観測することに用いられうる。
本発明の経鼻中枢神経系組織標識用組成物は、生きた生物個体の中枢神経系組織を、中枢神経系組織の露出や中枢神経系組織内または中枢神経系組織と連絡する組織内に染色剤を注入するという侵襲性の高い操作を要することなく、中枢神経系組織を標識することが可能である。従って、これらの識別を利用して、診断剤としての応用に用いることが出来る。
診断剤としては、特に限定されるものではないが、例えば、脳の機能を検査する診断剤や脳疾患の診断剤等に用いることが出来る。
診断対象の脳疾患は、特に限定されるものではないが、例えば、パーキンソン病、アルツハイマー病、ハンチントン舞踏病、運動ニューロン疾患、タウオパシー、皮質基底核変性症、うつ病、てんかん、偏頭痛、脊髄小脳変性症、脳腫瘍、脳出血、脳梗塞、などが挙げられる。
本発明の第四実施形態の具体例を以下に示す。
(1)脳機能イメージング・脳機能マッピング
本発明の経鼻中枢神経系組織標識用組成物は、従来のfMRIや近赤外光脳機能イメージング、あるいは内因性シグナルイメージング法の代替として、脳機能イメージングや脳機能マッピングを行うためのプローブとして用いることが出来る。本発明の経鼻中枢神経系組織標識用組成物は、神経細胞の軸索内部とシナプス間隙を移動することによって中枢神経系組織内部に移行する。一方、経鼻中枢神経系組織標識用組成物の蛍光特性は、相互作用する生体分子や、溶媒環境により変化する。そのため、この蛍光特性の変化を検出することにより、脳神経細胞の活動状態の変化を検出することが出来る。
(1)脳機能イメージング・脳機能マッピング

(2)嗅覚情報処理・嗅覚認知研究
嗅覚は、多くの動物が、その摂食行動、危険回避行動、繁殖行動など、生存に不可欠な行動を喚起するために利用している。嗅覚の受容体遺伝子群が発見されて以来、嗅覚の研究は急速に進んだが、「好き」なにおいへの誘引反応や、「嫌い」なにおいからの忌避反応といった、においで喚起される行動を支配する神経回路については未だ十分に解明されたわけではない。
嗅覚は、多くの動物が、その摂食行動、危険回避行動、繁殖行動など、生存に不可欠な行動を喚起するために利用している。嗅覚の受容体遺伝子群が発見されて以来、嗅覚の研究は急速に進んだが、「好き」なにおいへの誘引反応や、「嫌い」なにおいからの忌避反応といった、においで喚起される行動を支配する神経回路については未だ十分に解明されたわけではない。
本発明の経鼻中枢神経系組織標識用組成物は、動物の嗅覚神経回路を可視化し、その繋がりを明らかにするために用いることが出来る。におい物質と本発明の経鼻中枢神経系組織標識用組成物との共存により、染色性が変化する嗅覚神経回路を同定することにより、嗅球におけるにおい地図を描くことや、嗅覚情報伝達経路を同定することが可能となる。
(3)光増感剤(光線力学療法)
本発明の経鼻中枢神経系組織標識用組成物は、光増感剤として用いることもできる。光増感剤は、光活性化する光の照射によって活性化され、光増感剤それ自体またはいくつかの他の化学種(例えば酸素)が細胞毒性種に転換され、それによって光を照射した部位の標的細胞が殺傷されるか、またはそれらの増殖性潜在能力が減少する。
本発明の経鼻中枢神経系組織標識用組成物は、光増感剤として用いることもできる。光増感剤は、光活性化する光の照射によって活性化され、光増感剤それ自体またはいくつかの他の化学種(例えば酸素)が細胞毒性種に転換され、それによって光を照射した部位の標的細胞が殺傷されるか、またはそれらの増殖性潜在能力が減少する。
従って本発明の経鼻中枢神経系組織標識用組成物を用いると、中枢神経系組織の標識に基づいて、診断とともに治療も行うことができる。
本発明の第五の実施形態であるスクリーニング方法は、経鼻中枢神経系組織標識用組成物を用いて、in vivoで中枢神経系組織に作用する化合物を検出することを特徴とする。
本発明の経鼻中枢神経系組織標識用組成物は、生物個体、例えば、小型硬骨魚類であるゼブラフィッシュを用いて、生きたままの状態、所謂in vivoで、スクリーニング対象化合物の中枢神経系組織への経鼻移行性や安全性に関する評価に用いられうる。
ゼブラフィッシュは、米国及び英国では、近年、既にマウス及びラットに続く第3のモデル動物として認知されており、人と比較して全ゲノム配列が80%の相同性、遺伝子数もほぼ同じ、主要臓器・組織の発生・構造も良く似ていることが解明されてきている。各パーツ(心臓、肝臓、腎臓、消化管等の臓器・器官)が受精卵から分化して形成されていく過程が透明な体を通して観察できるのが特徴であるため、ゼブラフィッシュをモデル動物としてスクリーニングに用いることは特に好ましい。
「中枢神経系組織に作用する化合物を検出すること」とは、本発明の経鼻中枢神経系組織標識用組成物を用いて、調べたい化合物(スクリーニング対象化合物)を当該中枢神経系組織へ作用させた際の標識性の変化を測ることによって、中枢神経系組織に作用する化合物の有無や特性を検出することをいう。具体的に、一例として、スクリーニング対象化合物とゼブラフィッシュとを接触させて、本発明の経鼻中枢神経系組織標識用組成物を経鼻投与した時のゼブラフィッシュ脳染色性に及ぼす影響を観察するスクリーニング法が挙げられる。
スクリーニング対象化合物を接触させる方法としては、特に限定されるものではないが、スクリーニング対象化合物が水溶性の場合は、飼育水中にスクリーニング対象化合物を投与する方法、非水溶性の場合は、飼育水中にスクリーニング対象化合物を単独で分散させて投与する方法、または微量の界面活性剤やDMSOと共に投与する方法、ゼブラフィッシュの餌に混ぜて経口投与する方法、または、注射等による非経口投与する方法が挙げられる。好ましくは、簡便性から飼育水中にスクリーニング対象化合物を投与する方法が挙げられる。
前記スクリーニング対象化合物とは、生物化学的に作用のある化合物の総称を意味し、特に限定されるものではないが、例えば、医薬品、有機化合物、治療剤、治験薬、農薬、化粧品、環境汚染物質、内分泌攪乱物質、またはこれらの候補化合物等が挙げられる。
前記医薬品、治療薬またはこれらの候補化合物とは、中枢神経系組織への経鼻移行性に基づいて、例えばパーキンソン病、アルツハイマー病、ハンチントン舞踏病、運動ニューロン疾患、タウオパシー、皮質基底核変性症、うつ病、てんかん、偏頭痛、脊髄小脳変性症、脳腫瘍、脳出血、脳梗塞等を治療する医薬品またはその候補化合物、さらにはこれらの化合物の中枢神経系組織への経鼻移行を促進又は抑制する化合物等が挙げられる。
前記医薬品、治療薬またはこれらの候補化合物とは、中枢神経系組織への経鼻移行性に基づいて、例えばパーキンソン病、アルツハイマー病、ハンチントン舞踏病、運動ニューロン疾患、タウオパシー、皮質基底核変性症、うつ病、てんかん、偏頭痛、脊髄小脳変性症、脳腫瘍、脳出血、脳梗塞等を治療する医薬品またはその候補化合物、さらにはこれらの化合物の中枢神経系組織への経鼻移行を促進又は抑制する化合物等が挙げられる。
又、前記疾患等に起因する嗅覚障害を治療する医薬品またはその候補化合物等が挙げられる。
ゼブラフィッシュは、スクリーニングの目的に応じ、野生型ゼブラフィッシュに制限されず、各種疾患モデルのゼブラフィッシュを用いることが出来る。疾患モデルを用いた場合には、観察により新薬候補化合物の効果を見出し、疾患治療薬または疾患予防薬のスクリーニングに応用することが出来る。
また、スクリーニング対象化合物が共存する条件下に、本発明の経鼻中枢神経系組織標識用組成物を経鼻投与した時のゼブラフィッシュ脳染色性の経鼻染色速度を測定することで、スクリーニング対象化合物の経鼻中枢神経系組織への移行速度を評価することが可能になる。
また、本発明のスクリーニング方法は、小型硬骨魚類を用いることが出来る。本発明のスクリーニング方法で用いられる小型硬骨魚類としては、特に制限されるものではないが、例えば、ゼブラフィッシュ、フグ、金魚、メダカ、またはジャイアント・レリオ等が挙げられる。小型硬骨魚類は、マウス及びラット等と比較して、スピード面・コスト面で非常に優れているため好ましい。特に、ゼブラフィッシュはゲノムの解読がほぼ完了しており、また飼育及び繁殖が容易で流通価格も安く、受精後48~72時間で主要臓器・組織の基本構造が出来上がるため、好ましい。
(ヒトへの外挿性について)
本発明の経鼻中枢神経系組織標識用組成物は、ヒトへも適用可能である。ヒトへの外挿性は、ヒトと実験動物の中枢神経系組織の類似性、相違点を認識した上で全体的な近似によって確かめられる。以下に数例を示すが、これらに限定されるのではない。
(1)ヒト以外で、ヒトとヒト以外の生きた生物試料の中枢神経系組織を染色し、類似性を確認する。ヒト以外の生きた生物試料としては、マウス、ハムスター、ラット、モルモット、ウサギ、イヌ、豚、猫、サル等の哺乳類動物、ゼブラフィッシュ等の硬骨魚類等が挙げられる。
(2)前記ヒト以外の生きた生物試料の固定化組織切片を用いて、中枢神経系組織の染色性を確認し、生きた生物試料と同様の染色性があることを確認する。
(3)ヒトの固定化組織切片を用いて中枢神経系組織の染色性を確認する。
本発明の経鼻中枢神経系組織標識用組成物は、ヒトへも適用可能である。ヒトへの外挿性は、ヒトと実験動物の中枢神経系組織の類似性、相違点を認識した上で全体的な近似によって確かめられる。以下に数例を示すが、これらに限定されるのではない。
(1)ヒト以外で、ヒトとヒト以外の生きた生物試料の中枢神経系組織を染色し、類似性を確認する。ヒト以外の生きた生物試料としては、マウス、ハムスター、ラット、モルモット、ウサギ、イヌ、豚、猫、サル等の哺乳類動物、ゼブラフィッシュ等の硬骨魚類等が挙げられる。
(2)前記ヒト以外の生きた生物試料の固定化組織切片を用いて、中枢神経系組織の染色性を確認し、生きた生物試料と同様の染色性があることを確認する。
(3)ヒトの固定化組織切片を用いて中枢神経系組織の染色性を確認する。
以上の3点を確認することで、ヒトに対しても本発明の経鼻中枢神経系組織標識用組成物が適用できることを確認することが出来る。
ヒトへの外挿性確認の別の方法としては、本発明の経鼻中枢神経系組織標識用組成物を放射性標識して極微量をヒトの体内へ投与し、中枢神経系組織への局在を確かめることで確認することが出来る。この手法はマイクロドージング試験と呼ばれる。
また別の方法としては、(1)ヒト以外の生物試料の中枢神経系組織で、本発明の経鼻中枢神経系組織標識用組成物の標的生体分子または染色機構を同定する。(2)該標的生体分子または染色分子機構に相同なヒト生体分子を同定する。(3)ヒト以外の実験動物に遺伝子改変によって該ヒト生体分子または染色機構を導入する。(4)得られた実験動物を用いて、導入した生体分子または染色機構を介して染色されることを確認する。
ヒト以外の生物試料としては、特にゼブラフィッシュを好ましく用いることができる。中枢神経系組織は多くの脊椎動物の間で解剖学的、組織学的、生化学的に非常によく保存されており、発生についても同様である。ゼブラフィッシュを用いることで、マウスなどに較べて飼育コストが低く、用いる化合物の量も少量で済むなどの利点が高い。また、形態だけではなく、分子レベルでヒトとも高い相同性を持つことが示されている。以上のことから、ゼブラフィッシュを用いて本発明の経鼻中枢神経系組織標識用組成物のヒト外挿性を確認することが好ましい。
以下に実施例を挙げて、本発明をより詳細に説明するが、これらの実施例は、本発明のより一層の深い理解のために示される具体例であって、本発明は、これらの具体例に何ら限定されるものではない。なお、特に表示していない限りは「%」は質量基準である。
<実施例1>
1mg/mLの化合物1(前記式(5)で表わされた化合物)のDMSO溶液に蒸留水を加えて、化合物1の濃度が200ng/mLである染色液1を得た。24穴マルチプレート(IWAKI製)の任意のウェル中に、1mLの染色液1とゼブラフィッシュ7日胚(7dpf)の稚魚を入れ、1時間放置した。次に、ウェル中の染色液1を除去し、蒸留水1mLで置換する操作を10分以内に3回行った。最後の液交換後の時間経過を測りながら、予め定めた時間が経過した時点で、ウェルから稚魚を取り出し、スライドガラス上で5%低温融解アガロースゲルに埋没させて動きを制限し、蛍光実体顕微鏡(Leica社製 MZ16FA)を用いてゼブラフィッシュの側面及び頭頂部から中枢神経系組織の染色状態の観察を行った。また、共焦点顕微鏡(Zeiss社製 Pascal Exciter)を用いて頭頂部から中枢神経系組織の観察を行った。観察後のゼブラフィッシュは再びウェル中に戻し、またある時間経過後に観察を行うことを繰り返した。
1mg/mLの化合物1(前記式(5)で表わされた化合物)のDMSO溶液に蒸留水を加えて、化合物1の濃度が200ng/mLである染色液1を得た。24穴マルチプレート(IWAKI製)の任意のウェル中に、1mLの染色液1とゼブラフィッシュ7日胚(7dpf)の稚魚を入れ、1時間放置した。次に、ウェル中の染色液1を除去し、蒸留水1mLで置換する操作を10分以内に3回行った。最後の液交換後の時間経過を測りながら、予め定めた時間が経過した時点で、ウェルから稚魚を取り出し、スライドガラス上で5%低温融解アガロースゲルに埋没させて動きを制限し、蛍光実体顕微鏡(Leica社製 MZ16FA)を用いてゼブラフィッシュの側面及び頭頂部から中枢神経系組織の染色状態の観察を行った。また、共焦点顕微鏡(Zeiss社製 Pascal Exciter)を用いて頭頂部から中枢神経系組織の観察を行った。観察後のゼブラフィッシュは再びウェル中に戻し、またある時間経過後に観察を行うことを繰り返した。
その結果、図1A、図1B、図1Cに示す様に、時間経過と共に、最初は鼻腔内が、次に嗅球が、最後に中枢神経系組織の染色性が確認された。
又、図2に示す様に、中枢神経系組織における神経細胞の染色性が確認された。
<実施例2>
1mg/mLの化合物2(前記式(6)で表わされた化合物)のDMSO溶液に蒸留水を加えて、化合物2の濃度が500ng/mLである染色液2を得た。染色液1の代わりに染色液2を用いる他は、実施例1と同様の操作を行い、中枢神経系組織の観察を行った。
1mg/mLの化合物2(前記式(6)で表わされた化合物)のDMSO溶液に蒸留水を加えて、化合物2の濃度が500ng/mLである染色液2を得た。染色液1の代わりに染色液2を用いる他は、実施例1と同様の操作を行い、中枢神経系組織の観察を行った。
その結果、図3A、図3B、図3Cに示す様に、時間経過と共に、最初は鼻腔内が、次に嗅球が、最後に中枢神経系組織の染色性が確認された。又、図4A、図4A中の枠線内を拡大した図4B、さらには図4B中の枠線内を拡大した図4Cに示す様に、中枢神経系組織における神経細胞の染色性が確認された。
<比較例1>
1mg/mLのフルオレセインのDMSO溶液に蒸留水を加えて、フルオレセインの濃度が500ng/mLである染色液3を得た。染色液1の代わりに染色液3を用いる他は、実施例1と同様の操作を行い、中枢神経系組織の観察を行った。

その結果、図5に示す様に、染色性の時間経過に伴う、中枢神経系組織への移行性は観察されなかった。
<実施例3>
化合物3(前記式(7)で表わされた化合物)を特開2010-169677号に記載の方法と同様にして合成した。1mg/mLの化合物3のDMSO溶液に蒸留水を加えて、化合物3の濃度が500ng/mLである染色液4を得た。染色液1の代わりに染色液4を用いる他は、実施例1と同様の操作を行い、中枢神経系組織の観察を行った。
<実施例3>
化合物3(前記式(7)で表わされた化合物)を特開2010-169677号に記載の方法と同様にして合成した。1mg/mLの化合物3のDMSO溶液に蒸留水を加えて、化合物3の濃度が500ng/mLである染色液4を得た。染色液1の代わりに染色液4を用いる他は、実施例1と同様の操作を行い、中枢神経系組織の観察を行った。
その結果、図6A、図6B、図6Cに示す様に、時間経過と共に、最初は鼻腔内が、次に嗅球が、最後に中枢神経系組織の染色性が確認された。又、図7に示す様に、中枢神経系組織における神経細胞の染色性が確認された。
本発明により、生きている生物試料の中枢神経系組織を新しい投与経路で標識し、かつ中枢神経系組織の細胞の形態を高感度にイメージングすることが可能な経鼻中枢神経系組織標識用組成物が提供される。これは中枢神経系領域の研究や中枢神経系組織イメージング技術に必要不可欠な材料となる。また、中枢神経系疾患に関連する創薬開発において、経時的な評価を可能とする。さらに、ハイスループットで高精度なスクリーニングを低コストで行うことが可能となるため、新しい疾患の診断法や治療法の開発、更には中枢神経研究を飛躍的に発展させ、産業上及び実用化の上でもきわめて有効な基盤技術となる。
この出願は2009年12月25日に出願された日本国特許出願第2009-296329からの優先権を主張するものであり、その内容を引用してこの出願の一部とするものである。
Claims (5)
- 下記一般式(1)、または(2)のいずれかの化学式で表わされる化合物のうち少なくとも1種を有効成分とする、鼻腔内投与により生体の嗅上皮から嗅球を経て中枢神経系組織を標識することを特徴とする経鼻中枢神経系組織標識用組成物。
[一般式(1)中、R1は、アルキル基を表し、R3~R6は、各々独立して水素原子、アルキル基、アリール基、アルコキシ基、カルボン酸基、スルホン酸基、ヘテロ環基、アミノ基、またはハロゲン原子を表し、R5とR6は互いに結合して環を形成しても良い。X1 -は陰イオン性基を表す。Aは下記一般式(3)または(4)を表す。]

R11~R15は、各々独立して、水素原子、または、アルキル基を表し、X2 -は陰イオン性基を表す。]。
[一般式(3)中、R2は、アルキル基を表し、R7~R10は、各々独立して水素原子、アルキル基、アリール基、アルコキシ基、カルボン酸基、スルホン酸基、ヘテロ環基、アミノ基、またはハロゲン原子を表し、R7とR8は互いに結合して環を形成しても良い。
一般式(4)中、Yはアルキル基を表す。*は結合部位を示す。] - 前記化合物が蛍光性化合物であることを特徴とする請求項1に記載の経鼻中枢神経系組織標識用組成物。
- 前記化合物が放射性核種で標識されていることを特徴とする請求項1または請求項2に記載の経鼻中枢神経系組織標識用組成物。
- 請求項1乃至3のいずれか1つに記載の経鼻中枢神経系組織標識用組成物を用いて生体の嗅上皮から嗅球を経て中枢神経系組織を染色することを特徴とする中枢神経系組織標識方法。
- 請求項1乃至3のいずれか1つに記載の経鼻中枢神経系組織標識用組成物を用いて、in
vivoで中枢神経系組織に作用する化合物を検出することを特徴とするスクリーニング方法。
