Abstract

本発明は、留置後の内膜形成を促進し、内膜形成されるまでの間に抗血栓性を維持できる、長期開存可能な人工血管を提供することを目的としている。本発明は、極細繊維を含む繊維層の内側に、繊維径が10nm以上3μm以下の極細繊維からなる極細繊維層を備える筒状織物であり、ヘパリン以外の、高分子鎖を有する抗トロンビン剤が前記高分子鎖を介して前記極細繊維と共有結合され、37℃における繊維表面のトロンビン活性阻害比率が60%以上である、人工血管を提供する。

Description

抗血栓性を有する人工血管
 本発明は、損傷等を受けた血管の再建、修復又は代替に使用される人工血管に関する。
 高齢化やメタボリックシンドロームの人口増加によって、動脈硬化を煩う患者の数が増加している。動脈硬化は動脈壁の異常であり、血液の高血糖状態や高脂質状態が血管壁を変性させ、血管壁が脆弱になったり、肥厚したり、あるいは石灰化して硬く脆くなる。このような血管の変性は体中の血管のいたる所で起こるが、特に末梢の血管ではその影響が著しい。
 このような変性した血管の治療には、従来よりカテーテルを使ったバルーン拡張術やステント留置術などの低侵襲な血管内治療が行われているほか、損傷した血管を自己の血管又は人工血管で置換する外科手術が行われている。
 しかしながら、人工血管を留置した際に生体はこれを異物として認識し、人工血管の血液接触面では血液凝固反応が進行して血栓が形成される。
また、生体の血管は血液と接触する面に血管内皮細胞を有する内膜があり、血栓形成を阻害する役割を担っているが、留置された人工血管でも、血管内皮細胞が人工血管の血液接触面を被覆して内膜を形成する。しかしながら、この内膜が、内皮細胞に被覆されるまでの間は、人工血管は異物として認識されるため、内膜が形成されるまで血栓形成を防ぐ手段が必要となる。特に、細径の人工血管が使用される部位は血流量が少ないため血栓が沈着しやすく、少量の血栓でも血管を塞ぎ易い。細径の人工血管は長期的な成績が良好でなく臨床使用できるものが無いのが現状である。
 これらの問題を解決するため、従来から早期の内膜形成と抗血栓性の付与が、人工血管の開発において重要なポイントであった。
 内膜形成を促進する方法として、細胞の増殖因子や誘導因子を人工血管に担持させるほかに、人工血管そのものの構造素材としてポリエステルなどの繊維の織物、編物あるいは不織布が使用されてきた。とりわけ10ミクロン未満の極細繊維を含有させた場合には、極細繊維のサイズあるいは繊維間隙のサイズが細胞の増殖や浸潤に適していることが知られている(特許文献1、2、3)。更に極細繊維は血小板の付着も促し、留置した際に血管壁から血液が漏れるのを防止する効果もあることが知られている(特許文献4)。
 また、人工血管に抗血栓性を付与する方法としては、従来からヘパリンを担持する方法が行われてきた。繊維自体にはヘパリンを担持する能力がないので、十分なヘパリンを担持させるために、ヘパリンを含んだ、生分解性ポリマーやゼラチンなどからなるゲルを充填させる方法や(特許文献5)、ヘパリンを共有結合で繊維表面に固定した人工血管が知られている(特許文献6)。
 一方、ヘパリン以外の物質を使って抗血栓性を付与する方法としては、抗トロンビン剤、あるいは抗トロンビン剤を含有するポリマーをγ線などの高エネルギー線を使って表面に結合させる方法が知られている(特許文献7、8)。

 しかしながら、特許文献5に記載の人工血管のように、繊維間隙を充填してしまう場合、細胞浸潤が妨げられて内膜が遅れるだけでなく、ゼラチンなどに血小板が付着して血栓形成を却って助長してしまうという問題がある。また、特許文献6に記載の人工血管のように、ヘパリンを共有結合で繊維表面に固定する場合、ヘパリンの分子量が大きい故に、表面に結合できる量には限度があり、長期的な効果がないという問題がある。更には、親水性が極めて高いヘパリンを繊維表面に結合させるため、細胞接着性が低下するという問題がある。
 さらに、特許文献7、8に記載されるように、抗トロンビン剤をγ線などの高エネルギー線により表面に結合する方法があるが、抗トロンビン剤が変性して活性が低下し、十分な抗血栓性能を発揮できないという問題がある。さらに、基材表面への固定化反応は水溶液中で抗トロンビン剤を表面に吸着させた状態でγ線照射することによって達成されるが、表面張力の高い水溶液は疎水性の高いポリエステル極細繊維の間隙には浸透せず、内部の繊維表面まで均一に表面処理できないため、表面処理されていない部分が血栓形成反応を誘導するという問題がある。また、親水性ポリマーはポリエステルに吸着しないため、抗トロンビン剤自体がポリエステルと親和性を持たない限り吸着は起こらず、表面固定化量を低下させる原因にもなる。更に、抗トロンビン剤が、スペーサーとなる親水性ポリマーを介さずに表面に結合するため、空間的な自由度が無く、トロンビンと結合が制限されるという問題もある。
 このように、従来の人工血管は、細胞親和性と抗血栓性との両立が達成されておらず、特に、臨床で長期使用に耐えうる細径の人工血管は未だに世の中に無い。
 そこで本発明は、留置後の内膜形成を促進し、内膜形成されるまでの間に抗血栓性を維持できる、長期開存可能な人工血管を提供することを目的とする。
 本発明者らは上記課題を解決するため鋭意検討した結果、ヘパリン以外の、高分子鎖を有する抗トロンビン剤が高分子鎖部分を介して極細繊維に共有結合されることによって、極細繊維からなる微細な構造を保持したまま抗血栓性を付与できること、すなわち細胞親和性と抗血栓性の両立が実現できることを見出した。
 すなわち、本発明は、以下の(1)~(12)の人工血管を提供するものである。
(1)極細繊維を含む繊維層の内側に、繊維径が10nm以上3μm以下の極細繊維からなる極細繊維層を備える筒状織物であり、ヘパリン以外の、高分子鎖を有する抗トロンビン剤が前記高分子鎖を介して前記極細繊維と共有結合され、37℃における繊維表面のトロンビン活性阻害比率が60%以上である、人工血管。
(2)上記抗トロンビン剤の分子量は、3000以下である(1)記載の人工血管。
(3)120mmHgにおける透水性能が100mL/cm/min以上4000mL/cm/min未満である(1)又は(2)記載の人工血管。
(4)37℃の条件下、該人工血管1g当たり10mLの生理食塩水に24時間抽出した後における抽出液中のトロンビン活性阻害比率は、5%未満である、(1)~(3)のいずれか記載の人工血管。
(5)上記抗トロンビン剤は、グアニジノ基、グアニド基又はアミジノ基を有する、(1)~(4)のいずれか記載の人工血管。
(6)上記高分子鎖は、ポリアルキレングリコール、ポリビニルアルコール又はポリビニルピロリドンからなる高分子構造から選択される、(1)~(5)のいずれか記載の人工血管。
(7)上記抗トロンビン剤は、下記の化学式(I)~(IV)から選択される、(1)~(6)のいずれか記載の人工血管。(nは1~500までの整数とする。)

(8)上記繊維層は、上記極細繊維と、総繊度が1~60デシテックスのマルチフィラメントと、からなる、(1)~(7)のいずれか記載の人工血管。

 本発明によれば、留置後の内膜形成を促進し、内膜形成されるまでの間に抗血栓性を維持できる、長期開存可能な人工血管を提供することができる。

 本発明でいう極細繊維とは、繊維径が10nm以上、3μm以下の繊維を指す。極細繊維を用いた人工血管は、繊維が極細であるが故に生体細胞が付着するに適した足場の数が著しく増大し、細胞の浸潤にも優れ、内膜形成が極めて早期にかつ良好に行われるだけでなく、血液の漏出も著しく低いという特徴がある。
 極細繊維のみでは血圧や組織の動きに追従できる強度が発揮できないため、本発明の人工血管の繊維構造は、図1に示すように、太い繊維で構成された粗い織目、編目等で形成された基本組織の空隙に極細繊維1が散在する繊維層2と、繊維層2の内側に、極細繊維1からなる極細繊維層3を備えた繊維構造となっている。本発明の人工血管は、この繊維構造を筒状にすることで形成されている。
 粗い織目、編目等による基本組織の空隙に極細繊維が散在する繊維構造を形成する製法として、一般的な極細繊維の製法を採用することができ、基本組織の強度に適したサイズの繊維と共に、海島構造等を有する多成分系繊維を用いて織り、編み、組紐や不織布等に加工した後、アルカリなどを用いて多成分系繊維内の一部の繊維の海構造を溶解することで、極細化処理して基布中の極細繊維及び極細繊維層を作ることが好ましい。
 更に、極細化処理を行った後に、ウォータージェットやエアジェットなどで、基本組織に極細繊維を交絡させることによってより細胞にとって好ましい間隙構造が達成される。また、細胞親和性をより効果的に発揮する手段として、例えばヤスリ棒で血液接触面をこすって毛羽立たせる方法によって、血液接触面に極細繊維層をより形成させることができる。
 繊維素材としては、生体適合性を有するポリマーであれば特に限定されるものではない。例えば、ポリエステル、ポリエチレン、ポリテトラフルオロエチレン、ポリウレタン、ポリアミド、ナイロン等を用いることができる。これらの繊維素材の中でも、従来より人工血管の素材として臨床使用され、強度に優れる点でポリエステル、特にポリエチレンテレフタレートが好ましい。
 繊維の形態としては、紡績糸、マルチフィラメント糸、モノフィラメント糸、フィルム割繊糸といった如何なる形態でもよいが、強度や物性の均一性、及び柔軟性から、マルチフィラメント糸が優れている。また、無撚りであってもよいが、撚糸であっても問題は無い。ある程度のケン縮加工が施されていてもよい。
 繊維の総繊度は1~60デシテックス(Dtex)であることが好ましく、1~40デシテックスであることがより好ましい。総繊度の下限はより好ましくは5デシテックス以上であり、最も好ましくは10デシテックス以上である。総繊度の上限はより好ましくは35デシテックス以下であり、最も好ましくは25デシテックス以下である。1デシテックス以上であれば、基布に必要な強力を維持することができ、40デシテックス以下であれば、基布の厚みを低減することができる。
 繊維の単糸繊度は0.5~10デシテックス(Dtex)であることが好ましく、0.5~3.0デシテックスであることがより好ましい。単糸繊度の下限はより好ましくは1デシテックス以上であり、単糸繊度の上限はより好ましくは2デシテックス以下である。3デシテックス以上であると、柔軟性が損なわれ、0.5デシテックス以下であると、加水分解速度が速く、強度劣化の問題が生じる。
 また、人工血管を形成する筒状織物において、布帛の形態は、寸法安定性と強度に優れる点で織物を用いる。
 また、繊維表面に固定化する抗トロンビン剤の量を拡大させるためには、上記布帛の一部に極細単糸繊度マルチフィラメント糸を配することも効果的で、この極細単糸繊度マルチフィラメント糸の単糸繊維直径は10nm~20μmであることが好ましく、10nm~3μmであることがより好ましく、0.8~1.2μmであることが最も好ましい。
 人工血管の繊維層及び極細繊維層における、繊維間隙のサイズと量は、120mmHgの圧力下での透水性を指標として表すことができ、100mL/cm/min以上4,000mL/cm/min以下であることが好ましい。人工血管の血液接触面に安定した血管内皮細胞層を含む内膜が形成されるためには、それを下支えするための血管平滑筋や線維芽細胞を主体とする細胞層が重要であり、この細胞層中の細胞は、血管表面上を移動する血管内皮細胞と共に、繊維間隙を通って吻合部から内部へと侵入する。また、血管内皮細胞は吻合部から浸潤するだけでなく、人工血管外壁から繊維間隙を通して侵入してきた毛細血管が人工血管内壁で開通した部位からも浸潤する。
 このことから、繊維間隙は、100mL/cm/min以上であると、細胞や毛細血管が繊維層の内部に浸潤することによる内膜形成が起きやすくなるため好ましい。また、繊維間隙は、4,000mL/cm/min以下であると、細胞の偽足が繊維層の内部に届きやすくなり、間隙を塞いで漏血を防止するようになるため好ましい。
 本発明の人工血管のサイズは特に限定されるものではないが、内径が1mm以上、10mm未満の細径の人工血管において最も効果的である。
 本発明は、ヘパリン以外の、高分子鎖を有する抗トロンビン剤が、高分子鎖を介して基本組織を構成する繊維及び極細繊維の表面に共有結合することによって、細胞親和性と抗血栓性の両立を実現した。
 ヘパリン以外の、高分子鎖を有する抗トロンビン剤の中でも特に、低分子量の抗トロンビン剤を使用することが好ましい。具体的には、分子量が3000以下の抗トロンビン剤が好ましい。
 抗トロンビン剤として、一般的に知られているヘパリンは分子量が30,000~35,000ダルトンと大きな分子であるため、表面に固定化できる量には限度がある。ヘパリンよりも分子量の小さい低分子量ヘパリンも臨床使用されているが、低分子ヘパリンでさえも4,000~6,000の分子量であり、合成抗トロンビン物質の分子量の約10倍である。ヘパリンは、アンチトロンビンIIIおよびトロンビンと結合して初めてトロンビンの活性を阻害でき、アンチトロンビンIIIおよびトロンビンとの結合部位が分子内に別々に存在するため、これらの結合サイトを最適な配置で表面に固定化するのは極めて制御が難しく、表面固定化の反応効率が悪い原因にもなっている。また、ヘパリンの抗トロンビン活性そのものは、合成抗トロンビン物質の10分の1程度で、そもそもの活性が低い。更に、ヘパリンに対して過剰なアレルギーを示す、ヘパリン起因性血小板減少症の患者が世の中に少なからず存在することも知られており、社会的、倫理的な理由でヘパリンが全く自由に使用できるわけではない。
 本発明に用いる高分子鎖を有する抗トロンビン活性物質としては、グアニジノ基、グアニド基又はアミジノ基を有する物質であることが好ましく、下記一般式の(I)~(IV)から選ばれる物質がより好ましい。

 これらの高分子鎖を有する抗トロンビン剤は、高分子鎖末端の官能基を介して共有結合で繊維表面に固定化されるため、溶出することが無く、表面で長期的な効果を持続することができる。抗トロンビン活性を維持しつつ、抗トロンビン剤を繊維表面に共有結合で固定化する方法としては、抗トロンビン剤の活性部位と離れた、例えば高分子鎖を挟んだ箇所に、固定化反応に用いる反応性官能基を導入して誘導体とし、縮合反応、付加反応やグラフト重合反応などの化学反応で固定化するのが最も好ましい。抗トロンビン剤と繊維を共存させた状態でγ線、電子線などの高エネルギー線を照射する方法や、繊維をプラズマ処理した後に抗トロンビン剤と接触させる方法などは、反応性が高いラジカル種が活性部位を失活させたり、抗トロンビン剤が最大活性を発揮できるように配向性を制御できないために使用できない。
 また、抗トロンビン剤を固定する際には、固定部位を活性部位とできるだけ離し、更に固定化後の自由度を確保するために高分子鎖を介して反応性官能基を導入し固定化する方法が好ましい。また、高分子鎖は、親水性を有していることが好ましい。親水性の高分子鎖の構造は限定されるものではないが、親水性の血液中で特に効力を発揮するためには、ポリエチレングリコール(PEG)、ポリプロピレングリコール(PPG)、ポリエチレングリコール/ポリプロピレングリコール共重合体(PEG-PPG)などのポリアルキレングリコール、ポリビニルアルコール(PVA)、ポリビニルピロリドン(PVP)からなる高分子構造から選択されることが好ましい。親水性の高分子鎖の重合度nは特に限定されるものではないが、高すぎると親水性が強くなり細胞接着性が低下することから、1以上500以下であることが好ましい。
 本発明の人工血管が有する抗血栓性及び細胞親和性について、透水性、抽出液中のトロンビン活性阻害比率、繊維表面のトロンビン活性阻害比率、血小板付着率、細胞接着率及び血栓付着の測定を用いて示す。それぞれの測定方法は以下に示す。
<透水性>
 人工血管から無作為に2箇所をサンプリングし、各サンプルに対し下記方法で2回測定して、その相加平均を求める。人工血管を軸方向に沿って切り裂き、1cm角のサンプル断片を作成した。直径0.5cmの打ち抜きをした直径4cmのドーナッツ状パッキン2枚で、打ち抜き部分以外に通液のないよう、1cm角の織物試料を挟む。これを円形ろ過フィルター用ハウジングに収納する。この円形ろ過フィルターに温度25℃の逆浸透膜ろ過水をサンプル断片が十分含水するまで2分以上通液する。温度25℃、ろ過差圧120mmHgの条件下で、逆浸透膜ろ過水の外圧全ろ過を30秒間行い、直径1cmの部分を透過する水の透過量(mL)を測定する。透過量は小数第1位を四捨五入して求める。その透過量(mL)を単位時間(min)及びサンプル断片の有効面積(cm)あたりの値に換算して、圧力120mmHgにおける透水性能を測定する。
 抗トロンビン剤固定化後の人工血管について、抗トロンビン剤の溶出がどれだけおきるかを調べる方法としては、抽出液中のトロンビン活性阻害比率を以下の方法により測定することができる。この抽出液中のトロンビン活性阻害比率について、抽出液の37℃におけるトロンビン活性阻害比率は低い程よく、5%未満であることが好ましく、1%未満であることがより好ましい。
<抽出液中のトロンビン活性阻害比率測定方法>
 人工血管を横断面方向に輪切りにしたリング状サンプル1gを元の血管の長軸方向0.1gずつ10個の小片に分割し、サンプル1g当たり10mLの生理食塩水で37℃、24時間の条件で抽出する。サンプルを含まない生理食塩水とサンプル抽出液10μLに対して、0.1U/mLトロンビン(Haematologic Technologies社)水溶液0.5mL及び200μMのS2238(積水メディカル社)水溶液0.5mLを添加する。37℃で45分静置後、405nmの吸光度をマイクロプレートリーダー(コロナ電気株式会社、MTP-300)で測定し、パラニトロアニリンの波長316nmにおけるモル吸光係数(1.27×10mol-1・L・cm-1)を用いて単位時間あたりのS2238分解量、すなわち分解速度を算出した。以下の式1に示すように、サンプルを含まない生理食塩水の分解速度を100としたとき、抽出液の分解速度の割合を求め、37℃におけるトロンビン活性阻害比率を算出する。この式1を用いて抽出液中のトロンビン活性阻害比率とする。
 トロンビン活性阻害比率(%)=(1-抽出液の分解速度/生理食塩水の分解速度)×100・・・(式1)
 抗トロンビン剤固定化後の人工血管について、繊維表面の抗トロンビン能は、以下の繊維表面のトロンビン活性阻害比率の測定方法により測定することができる。この繊維表面の抗トロンビン活性阻害比率について、トロンビン活性阻害比率は高い程よく、60%以上であることが好ましく、80%以上であることがより好ましい。
<繊維表面のトロンビン活性阻害比率測定方法>
 人工血管を横断面方向に輪切りにしたリング状サンプル1gを元の血管の長軸方向0.1gずつ10個の小片に分割し、サンプル1g当たり0.1U/mLトロンビン(Haematologic Technologies社)水溶液5mL及び200μMのS2238(積水メディカル社)水溶液0.5mLを添加する。37℃で45分静置後、405nmの吸光度をマイクロプレートリーダー(コロナ電気株式会社、MTP-300)で測定し、パラニトロアニリンの波長316nmにおけるモル吸光係数(1.27×10mol-1・L・cm-1)を用いて単位時間あたりのS2238分解量、すなわち分解速度を算出した。この分解速度から、以下の式2に示すように、サンプルを含まない生理食塩水の分解速度を100としたとき、繊維表面の分解速度の割合を求め、37℃におけるトロンビン活性阻害比率を算出する。この式2を用いて繊維表面のトロンビン活性阻害比率とする。
 トロンビン活性阻害比率(%)=(1-繊維表面の分解速度/生理食塩水の分解速度)×100・・・(式2)
 抗トロンビン剤固定化後の人工血管について、繊維表面の血小板付着率は、下記の繊維表面の血小板付着率測定方法により測定できる。この繊維表面の血小板付着率は低い程よく、20%未満であることが好ましい。
<繊維表面の血小板付着率測定方法>
 人工血管を軸方向に沿って切り裂き、打抜ポンチで直径12mmの円板サンプルを打ち抜く。細胞培養用の24ウェル・マイクロプレート(住友ベークライト社)のウェルに血液接触面を上にして1枚入れ、上から肉厚3mmの金属パイプ状錘を乗せる。別調整した多血小板血漿を、血小板数が1ウェル当たり10個程度になるように添加する。37℃で2時間静置後、サンプルを取りだし、PBS(-)(ニッスイ社)でリンスした後にLDH Cytotoxicity Detection kit(タカラバイオ社)に記載のプロトコール通りに、血小板を破壊して生成したLDHの活性を測定する。別に作成した検量線から付着した血小板数を求める。以下の式3に示すように、サンプル断片に接触させる前の多血小板血漿の血小板数に対する、接触した血小板数の割合を求め、血小板付着率とする。
 血小板付着率(%)=(接触後の血小板付着数/多血小板血漿の血小板数)×100・・・(式3)
<細胞接着性>
 人工血管を軸方向に沿って切り裂き、打抜ポンチで直径12mmの円板サンプルを打ち抜く。細胞培養用の24ウェル・マイクロプレート(住友ベークライト社)のウェルに1枚入れ、上から肉厚3mmの金属パイプ状錘を乗せる。10%FCS含有DMEM培地に懸濁した正常ヒト臍帯静脈内皮細胞(タカラバイオ社)を1ウェル当たり10個になるように添加する。37℃で12時間静置後、サンプルを取りだし、PBS(-)(ニッスイ社)でリンスした後に酵素処理で細胞を剥がした後、MTTアッセイキット(フナコシ社)を用いて剥離した細胞数を測定する。以下の式4に示すように、サンプルに播種した細胞数に対する、接着した細胞数の割合を求め、細胞接着率とする。
 細胞接着率(%)=(接着した細胞数数/播種した細胞数)×100・・・(式4)
<血液循環における血栓付着>
 人工血管を長さ4cmに切り、人工血管と同じ内径のポリ塩化ビニル製チューブ32cmと接続した。ヘパリンを終濃度0.5IU/mLとなるよう添加したヒト新鮮血4.5mLをチューブ内に導入し、ただちに両端を閉じてループとした。これを予め37℃に調節した恒温恒湿乾燥機内で、回転速度14rpmに調節したローテーターに貼り付けた枠へ固定し、120分間回転させた。ループを取り出し、ポリ塩化ビニル製チューブを切断して血液を除去しPBS(-)(ニッスイ社)でリンスした後、人工血管内の血栓形成の有無を定量する。試験はN=3で行う。陰性対照として、ヒト新鮮血の代わりにPBS(-)を用いて同様の試験を行う。試験前と、血液を除去しリンス後の4cmの人工血管の乾燥重量をそれぞれ測定し、その差を血栓重量とし、平均値及び標準偏差を算出する。試料の平均値が、陰性対照の(平均値+3×標準偏差)以上であれば「+」、未満であれば「-」と判定する。循環中に人工血管を通して血液が漏れた場合には、量の如何を問わず「漏出」として試験を止める。
 以下、本発明の人工血管の具体的な実施例について詳細に説明する。
(実施例)
 経糸55Dtex-48fのポリエチレンテレフタレート、緯糸に245Dtex-40fの高分子配列体繊維を用いて平織組織で筒状織物を作製した。この時用いた高分子配列体繊維は海成分ポリスチレン20部、島成分ポリエチレンテレフタレート80部で島数36/fであった。この筒状織物を80℃の水酸化ナトリウム水溶液で十分に処理した後、トルエン中に浸漬し、次いで起毛機で起毛しさらにウォータージェットパンチを行った。
 上記処理後の筒状織物を0.5%水酸化ナトリウム水溶液で処理した後、次いで5%過マンガン酸カリウムで酸化処理した。続いて、一般式(I)~(IV)が有する親水性の高分子鎖部分の末端にアミノ基を導入した一般式(V)~(VIII)化合物をそれぞれ1~50mg/mL溶液中に筒状織物を浸漬し、0.1%カルボジイミドの存在下で縮合反応させ、生理食塩水により洗浄したものを人工血管として用いる抗血栓筒状織物とした。

 それぞれの抗血栓筒状織物の、透水性、抽出液中のトロンビン活性阻害比率、繊維表面のトロンビン活性阻害比率、血小板付着率、細胞接着率及び血栓付着の測定の性能評価結果を表1に示す。
 ここで、一般式(V)の抗トロンビン剤1mg/mL中で処理した抗血栓筒状織物を実施例1、一般式(V)の抗トロンビン剤2mg/mL中で処理した抗血栓筒状織物を実施例2、一般式(V)の抗トロンビン剤5mg/mL中で処理した抗血栓筒状織物を実施例3、一般式(V)の抗トロンビン剤10mg/mL中で処理した抗血栓筒状織物を実施例4、一般式(V)の抗トロンビン剤20mg/mL中で処理した抗血栓筒状織物を実施例5、一般式(V)の抗トロンビン剤50mg/mL中で処理した抗血栓筒状織物を実施例6、一般式(VI)の抗トロンビン剤50mg/mL中で処理した抗血栓筒状織物を実施例7、一般式(VII)の抗トロンビン剤50mg/mL中で処理した抗血栓筒状織物を実施例8、一般式(VIII)の抗トロンビン剤50mg/mL中で処理した抗血栓筒状織物を実施例9とした。
(比較例)
 実施例1で得たものと同様の筒状織物を、それぞれ一般式(I)~(IV)の抗トロンビン剤50mg/mL水溶液に浸漬し、その状態でコーガアイソトープにて5kGyγ線照射を行った。Triton-X100と水で洗浄して抗血栓筒状織物とした。
ここで、一般式(I)の抗トロンビン剤50mg/mL中でγ線照射した抗血栓筒状織物を比較例1、一般式(II)の抗トロンビン剤50mg/mL中でγ線照射した抗血栓筒状織物を比較例2、一般式(III)の抗トロンビン剤50mg/mL中でγ線照射した抗血栓筒状織物を比較例3、一般式(IV)の抗トロンビン剤50mg/mL中でγ線照射した抗血栓筒状織物を比較例4とした。それぞれの抗血栓筒状織物の、透水性、抽出液中のトロンビン活性阻害比率、繊維表面のトロンビン活性阻害比率、血小板付着率、細胞接着率及び血栓付着の測定の性能評価結果を表1に示す。

 経糸、緯糸ともに55Dtex-48fのポリエチレンテレフタレートを用いて平織組織で筒状織物を作製した。一般式(I)の抗トロンビン剤50mg/mLとなる水溶液に浸漬し、その状態でコーガアイソトープにて5kGyγ線照射を行った。Triton-X100と水で洗浄して抗血栓筒状織物(比較例7)とした。抗血栓筒状織物の、透水性、抽出液中のトロンビン活性阻害比率、繊維表面のトロンビン活性阻害比率、血小板付着率、細胞接着率及び血栓付着の測定の性能評価結果を表1に示す。
 一般式(IX)の化合物をジメチルホルムアミド溶液に、4N塩酸/1,4-ジオキサン、(東洋化成社)を滴下し反応させ、一般式(IX)の塩酸塩を得た。該塩酸塩のジメチルホルムアミド溶液に、ジシクロヘキシルカルボジイミドおよび4-ヒドロキシベンゾトリアゾールをそれぞれ添加し、更にポリエーテル変性シリコーン(X-22-3939A:信越シリコーン社)を添加して反応させた。反応液を透析チューブ(スペクトラポアRC ポア6 MWCO=1000)に入れ、反応液の10倍体積の蒸留水中で透析した。透析後の反応液を濾過し、濾液の溶媒を除去し、乾燥して親水性高分子化合物を得た。得られた親水性高分子化合物50mg/mLとなる水溶液に浸漬し、その状態でコーガアイソトープにて5kGyγ線照射を行った。Triton-X100と水で洗浄して抗血栓筒状織物(比較例8)とした。抗血栓筒状織物の、透水性、抽出液中のトロンビン活性阻害比率、繊維表面のトロンビン活性阻害比率、血小板付着率、細胞接着率及び血栓付着の測定の性能評価結果を表1に示す。

 表1に示すとおり、抗トロンビン剤を縮合反応で固定した場合には人工血管の繊維表面の抗トロンビン活性が高く、循環しても血栓が形成されなかったのに対し、γ線照射により固定した場合には表面抗トロンビン活性が低く、血栓が形成された。又、透水性が高すぎると循環において漏血した。更に、スペーサー無く固定化した場合にも血栓が形成された。抗トロンビン剤を縮合反応で固定した場合は抗トロンビン剤が繊維の微細構造の深くまで表面処理され血栓形成抑制効果が高い。
 本発明は、抗血栓性と細胞親和性を両立し、留置後の内膜形成を促進し、内膜形成されるまでの間に抗血栓性を維持し、長期開存可能な人工血管として用いることができる。

Claims (12)

  1.  極細繊維を含む繊維層の内側に、繊維径が10nm以上3μm以下の極細繊維からなる極細繊維層を備える筒状織物であり、
     ヘパリン以外の、高分子鎖を有する抗トロンビン剤が前記高分子鎖を介して前記極細繊維と共有結合され、
     37℃における繊維表面のトロンビン活性阻害比率が60%以上である、人工血管。
  2.  前記抗トロンビン剤の分子量は、3000以下である請求項1記載の人工血管。
  3.  120mmHgにおける透水性能が100mL/cm/min以上4000mL/cm/min未満である請求項1又は2記載の人工血管。
  4.  37℃の条件下、該人工血管1g当たり10mLの生理食塩水に24時間抽出した後における抽出液中のトロンビン活性阻害比率は、5%未満である、請求項1~3のいずれか一項記載の人工血管。
  5.  前記抗トロンビン剤は、グアニジノ基、グアニド基又はアミジノ基を有する、請求項1~4のいずれか一項記載の人工血管。
  6.  前記高分子鎖が、ポリアルキレングリコール、ポリビニルアルコール又はポリビニルピロリドンからなる高分子構造から選択される、請求項1~5のいずれか一項記載の人工血管。
  7.  前記抗トロンビン剤は、下記の化学式(I)~(IV)から選択される、請求項1~6のいずれか一項記載の人工血管。(nは1~500までの整数とする。)

  8.  前記繊維層は、前記極細繊維と、総繊度が1~60デシテックスのマルチフィラメントと、からなる、請求項1~7のいずれか一項記載の人工血管。
  9.  前記マルチフィラメントを構成する単糸の繊度は、0.5~10.0デシテックスである、請求項8記載の人工血管。
  10.  血小板付着率が20%未満である、請求項1~9のいずれか一項記載の人工血管。
  11.  前記筒状織物は、ポリエステル繊維からなる、請求項1~10のいずれか一項記載の人工血管。
  12.  前記筒状織物の内径は、1mm以上10mm未満である、請求項1~11のいずれか一項記載の人工血管。
